As an autosomal dominant genetic disorder, neurofibromatosis type 1 (NF1) results from loss-of-function variants in the tumour suppressor gene NF1 and predisposes patients to tumours. NF1 is one of the most common tumour predisposition syndromes and an extremely variable multisystem disorder. The incidence of NF1 is estimated to be 1 in 3000, but this may be underestimated. Life expectancy for patients with NF1 is at least 8 years shorter for both men and women. Most individuals with NF1 are diagnosed in childhood.
